Gene silencing by RNA Interference (RNAi) is a powerful research tool for studying gene function in mammalian cells. RNAi is a biological phenomenon by which double stranded RNA (dsRNA) specifically reduces gene expression of its corresponding gene. Potent inhibition of specific gene expression is experimentally achieved by the transfection of small interfering RNA (siRNA), which binds RISC complex and cause degradation of target complementary mRNA molecules in the cell. Therefore, successful, potent RNAi experimentation is dependent upon the highly efficient delivery of the siRNA into cells by transfection of stable and functional siRNA molecules.
Most DNA transfection reagents are incompatible with siRNA, sensitive to serum, or are capable of inducing cytotoxicity, which negatively affects gene expression studies. Transfection Reagents have been developed and optimized specifically for use with siRNA transfection and allow transfection in the presence of serum without lowering transfection efficiency or reducing cell viability. Ready-to-run transfection protocols eliminate the need for extensive siRNA optimization experiments. Altogen Biosystems kits are also compatible for transfection of microRNA (miRNA) molecules.

A549 (human lung carcinoma cells) |

A549 was derived in 1972 from the lung carcinomatous tissue of a 58-year-old Caucasian male. A549 cells are adenocarcinomic human alveolar basal epithelial cells and are squamous in nature and responsible for the diffusion of substances such as water and electrolytes, across the alveoli of lungs. This is a hypotriploid human cell line with the modal chromosome number of 66, occurring in 24 percent of cells. There were also frequent instances of cells with 64, 65, and 67 chromosome counts. A549 cells have been shown to synthesize lecithin with a high percentage of desaturated fatty acids via the cytidine diphosphocholine pathway. These cells are used in vitro model for a type II pulmonary epithelial cell model for drug metabolism. Immunoperoxidase staining shows that A549 cells are positive for keratin. This cell line is typically used to investigate a numerous respiratory ailments, such as viral infections that can cause asthma, tissue damage linked to asbestos exposure, and smoking-related emphysema.
