Human Osteosarcoma Cells / SaOS (Human Osteoblastic Cells)

Hypotriploid human cell line was initiated from an osteosarcoma in a 14 year -old male Caucasian. The modal chromosome number was 66 occurring in 44 percent of cells. The rate of cells with higher ploidies was two percent. Eighteen to 19 marker chromosomes were common to all cells. Osteosarcoma is a highly metastatic cancer, with a redundancy of autocrine loops. Human osteosarcoma cells release matrix degrading enzymes in response to chemokine activation. Many cell lines derived from an osteosarcoma produce high yields of interferon after superinduction with polyinosinic acid polycytidylic acid, cycloheximide, and actinomycin D.

SaOS -2 is a non-transformed cell line derived in 1973 from the primary osteosarcoma of an 11-year old Caucasian girl. It was determined in 1987 that SaOS -2 cells possess several osteoblastic features and could be useful as a permanent line of human osteoblast-like cells and as a source of bone-related molecules. SaOS-2 have well-documented characterization data, the possibility to obtain large amounts of cells in short time, and can be fully differentiated in a manner that the osteoblastic cells naturally do. SaOS-2 cells can deposit a mineralization-competent extracellular matrix which makes the cells an important model for investigating events associated with the late osteoblastic differentiation stage in human cells. It has also been shown that Saos-2 cells possess several osteoblastic features and could be useful as a permanent line of human osteoblast-like cells and as a source of bone-related molecules.
